Others have suggested that the cancer phenotype may be erased via cellular reprogramming. Several cancer cell lines have been reprogrammed previously to varying degrees; however, it remains unclear if cancer cells can be fully reprogrammed into induced pluripotent stem cells (iPSCs) that can form benign teratomas, and if cancer-specific epigenetic alterations are erased during this process. We investigated if mammary cancer stem cell (CSC), derived from Her2/Neu over-expressing transgenic mice, can be fully reprogrammed into iPSCs.

We also tested whether the reprogrammed CSCs lose or retain their potential to form tumors in vivo by means of becoming pluripotent. We show that the reprogrammed breast CSCs can be terminally differentiated into mammary epithelial cells, and that tumorigenicity is retained during differentiation in this microenvironment. Reprogramming of cancer cells is a promising new research approach may offer new possibilities in modeling cancer in vitro.
