Mutations in the tumor suppressor p53, which are detected in more than 50% of human tumors, result in not only loss of function as a transcription factor, but also gain of oncogenic activities (gain of function: GOF). Our recent findings suggest that mutant p53 is inherently unstable in normal tissues but get stabilized in tumors, leading to induction of the GOF activity. Intriguingly, downregulation of mutant p53 in cancer cells significantly reduces their malignant properties, providing the rationale to deplete mutant p53 as a strategy for cancer therapy. By performing high-throughput screening using ~9,000 compounds, we have identified a drug that specifically triggers degradation of p53 mutants, but has little effect on wild-type p53. Given that mutations in the p53 gene are found only in tumors in most cases, this drug could be used for molecular targeted therapy for a wide range of cancers.
