An epithelial program driven by KLF4 stoichiometry determines hallmarks of somatic cell reprogramming

Through the expression of Oct3/4, Sox2, Klf4, and c-Myc, somatic cells reprogram to induced pluripotent stem cells (iPSCs), yet the exact mechanisms remain to be elucidated. We employed a piggyBac transposon vector system to assess the cellular and molecular hallmarks of mouse somatic cell reprogramming, and revealed a KLF4 cDNA variation that ultimately affects KLF4 protein levels. As a result, high-KLF4 generates few, but fully pluripotent iPSCs. In contrast, low-KLF4 gives rise to mostly partially reprogrammed cells.

Using expression analysis at intermediate reprogramming stages, we observed a dominant epithelial gene expression profile in the high-KLF4 condition. We identified cell surface markers and confirmed their presentation by FACS. Using CRISPR/Cas9, we performed a focused screen for modulators of the mesenchymal-to-epithelial transition (MET) by gene knockout. We are now employing these markers to fractionate cells with the intention of identifying intermediate cell populations that contribute to “true” iPSCs. Beyond reprogramming, I will discuss how our findings have initiated an active branch of research into human epidermal development and genodermatoses.
